Brighter reporter genes from multimerized fluorescent proteins.
814 BioTechniques Vol. 39, No. 6 (2005) The discovery and development of the green fluorescent protein (GFP) has opened up new areas of biological research, particularly for the visualization of protein dynamics and gene expression in living cells and organisms (1,2). The development of wavelengthshifted variants of GFP has further expanded the impact on biological research. One of the fluoresc...
متن کاملSupercharged green fluorescent proteins as bimodal reporter genes for CEST MRI and optical imaging.
Superpositively charged mutants of green fluorescent protein (GFP) demonstrated a dramatically improved chemical exchange saturation transfer (CEST) MRI contrast compared to their wild type counterparts. The mutants +36 GFP and +48 GFP were successfully expressed in mammalian cells and retained part of their fluorescence, making them a new potential bimodal reporter gene.
متن کاملFluorescent Reporter Genes and the Analysis of Bacterial Regulatory Networks
The understanding of the regulatory networks controlling the adaptation of bacteria to changes in their environment is critically dependent on the ability to monitor the dynamics of gene expression. Here, we review the use of fluorescent reporter genes for dynamically quantifying promoter activity and other quantities characterizing gene expression. We discuss critical physical and biological p...
متن کاملThe Single T65S Mutation Generates Brighter Cyan Fluorescent Proteins with Increased Photostability and pH Insensitivity
Cyan fluorescent proteins (CFP) derived from Aequorea victoria GFP, carrying a tryptophan-based chromophore, are widely used as FRET donors in live cell fluorescence imaging experiments. Recently, several CFP variants with near-ultimate photophysical performances were obtained through a mix of site-directed and large scale random mutagenesis. To understand the structural bases of these improvem...
متن کاملFluorescent and Bioluminescent Reporter Myxoviruses
The advent of virus reverse genetics has enabled the incorporation of genetically encoded reporter proteins into replication-competent viruses. These reporters include fluorescent proteins which have intrinsic chromophores that absorb light and re-emit it at lower wavelengths, and bioluminescent proteins which are luciferase enzymes that react with substrates to produce visible light. The incor...
